Switch #:
SWTI000606
Switch type:
Binary
Switch subtype:
Physicochemical compatibility

Description:
Phosphorylation of S645 in Phosphatidylinositol 4-phosphate 5-kinase type-1 gamma (Pip5k1c) impedes binding to AP-2 complex subunit beta (Ap2b1), while dephosphorylation by calcineurin promotes binding. These phosphorylation and dephosphorylation events are important for the regulation of clathrin coat formation associated with synaptic vesicles.

Participants:
(1) Phosphatidylinositol 4-phosphate 5-kinase type-1 gamma (Pip5k1c)
(2) AP-2 complex subunit beta (Ap2b1)

Interactions
Interaction #1 Pip5k1c - Ap2b1

Interfaces
(1) ELM:TRG_AP2beta_CARGO_2 motif (633IYFPTDERSWVY644) in Phosphatidylinositol 4-phosphate 5-kinase type-1 gamma (Pip5k1c)
(2) Beta2-adaptin appendage, C-terminal sub-domain (826-936) in AP-2 complex subunit beta (Ap2b1)

Interaction Regulation
PTM-dependent Inhibition (Phosphorylation of S645 on Phosphatidylinositol 4-phosphate 5-kinase type-1 gamma (Pip5k1c)) of the Phosphatidylinositol 4-phosphate 5-kinase type-1 gamma (Pip5k1c) TRG_AP2beta_CARGO_2 motif - AP-2 complex subunit beta (Ap2b1) Beta2-adaptin appendage, C-terminal sub-domain interaction

Regulatory Enzymes for switch
