Switch #:
Switch type:
Switch subtype:
Altered binding specificity

Phosphorylation of Myc proto-oncogene protein (MYC) at S62 by Mitogen-activated protein kinase 1 (MAPK1) primes MYC for phosphorylation by Glycogen synthase kinase-3 beta (GSK3B), which targets MYC to the SCF ubiquitin ligase complex, F-box/WD repeat-containing protein 7 (FBXW7) that marks MYC for degradation.

(1) Myc proto-oncogene protein (MYC)
(2) Mitogen-activated protein kinase 1 (MAPK1)
(3) Glycogen synthase kinase-3 beta (GSK3B)
(4) F-box/WD repeat-containing protein 7 (FBXW7)

This is a sequential switch. The interactions in this switch take place in a defined order. The order is defined by numbers to the left of each interaction. More than one interaction can occur in each step, these interactions occur independently but are necessary for future step
1Interaction #1 MYC - MAPK1

Is mutually exclusive with

(1) MOD_ProDKin_1 motif (59PPLSPSR65) in Myc proto-oncogene protein (MYC)
(2) Protein kinase domain (25-313) in Mitogen-activated protein kinase 1 (MAPK1)

(1) The Fbw7 tumor suppressor regulates glycogen synthase kinase 3 phosphorylation-dependent c-Myc protein degradation.
