Switch #:
SWTI000287
Switch type:
Specificity
Switch subtype:
Altered binding specificity

Description:
Phosphorylation of T-cell surface glycoprotein CD3 epsilon chain (CD3E) by Lck (Tyrosine-protein kinase Lck (LCK)) during T cell activation switches the specificity of CD3E from SH3 domain containing proteins like Epidermal growth factor receptor kinase substrate 8-like protein 1 (EPS8L1) to SH2 domain containing proteins like Tyrosine-protein kinase ZAP-70 (ZAP70).

Participants:
(1) T-cell surface glycoprotein CD3 epsilon chain (CD3E)
(2) Epidermal growth factor receptor kinase substrate 8-like protein 1 (EPS8L1)
(3) Tyrosine-protein kinase ZAP-70 (ZAP70)

Interactions
Interaction #2 CD3E - ZAP70

Is mutually exclusive with Interaction #1 CD3E - EPS8L1

Interfaces
(3) LIG_TYR_ITAM motif (185NPDYEPIRKGQRDLYSGL202) in T-cell surface glycoprotein CD3 epsilon chain (CD3E)
(4) SH2 domain (10-87) and (163-239) in Tyrosine-protein kinase ZAP-70 (ZAP70)

Interaction Regulation
PTM-dependent Induction (Phosphorylation of Y188 and Y199 on T-cell surface glycoprotein CD3 epsilon chain (CD3E)) of the T-cell surface glycoprotein CD3 epsilon chain (CD3E) LIG_TYR_ITAM motif - Tyrosine-protein kinase ZAP-70 (ZAP70) SH2 domain interaction
