Switch #:
SWTI000530
Switch type:
Binary
Switch subtype:
Physicochemical compatibility

Description:
Binding of the BIR domain-binding motif of Caspase-1 (Dcp-1) to the BIR domains of Apoptosis 1 inhibitor (th) requires cleavage of Caspase-1 (Dcp-1) at D33, since this results in a functional neo N-terminal motif. BIR domains are found in Inhibitor of Apoptosis Proteins (IAPs) that suppress the activity of activated caspases, either by directly inhibiting caspase catalytic activity, or by targeting caspases for degradation by ubiquitin modification.

Participants:
(1) Caspase-1 (Dcp-1)
(2) Apoptosis 1 inhibitor (th)

Interactions
Interaction #1 Dcp-1 - th

Interfaces
(1) LIG_BIR_III_4 motif (33DAKGC37) in Caspase-1 (Dcp-1)
(2) Inhibitor of Apoptosis domain (47-111) in Apoptosis 1 inhibitor (th)

Interaction Regulation
PTM-dependent Induction (Proteolytic cleavage at D33 on Caspase-1 (Dcp-1)) of the Caspase-1 (Dcp-1) LIG_BIR_III_4 motif - Apoptosis 1 inhibitor (th) Inhibitor of Apoptosis domain interaction

Context
Switch localisation
ELM curated: cytosol, mitochondrion
References

(1) IAPs are functionally non-equivalent and regulate effector caspases through distinct mechanisms.
