Switch #:
SWTI000680
Switch type:
Binary
Switch subtype:
Physicochemical compatibility

Description:
Phosphorylation of Y1176 in the endocytotic motif of Neural cell adhesion molecule L1 (L1CAM) by Proto-oncogene tyrosine-protein kinase Src (SRC) abolishes binding to the AP-2 complex subunit mu (AP2M1) and thereby inhibits internalisation of Neural cell adhesion molecule L1 (L1CAM).

Participants:
(1) Neural cell adhesion molecule L1 (L1CAM)
(2) AP-2 complex subunit mu (AP2M1)

Interactions
Interaction #1 L1CAM - AP2M1

Interfaces
(1) TRG_ENDOCYTIC_2 motif (1176YRSL1179) in Neural cell adhesion molecule L1 (L1CAM)
(2) Adaptor complexes medium subunit family (159-435) in AP-2 complex subunit mu (AP2M1)

Interaction Regulation
PTM-dependent Abrogation (Phosphorylation of Y1176 on Neural cell adhesion molecule L1 (L1CAM)) of the Neural cell adhesion molecule L1 (L1CAM) TRG_ENDOCYTIC_2 motif - AP-2 complex subunit mu (AP2M1) Adaptor complexes medium subunit family interaction

Regulatory Enzymes for switch
Modifying enzymes for residue: Y1176: Proto-oncogene tyrosine-protein kinase Src (SRC)

Context
References

(1) L1 endocytosis is controlled by a phosphorylation-dephosphorylation cycle stimulated by outside-in signaling by L1.
