Switch #:
SWTI000319
Switch type:
Specificity
Switch subtype:
Motif hiding

Description:
Phosphorylation-induced binding of 14-3-3 protein beta/alpha (YWHAB) promotes cell surface expression of G-protein coupled receptor 15 (GPR15) by releasing the receptor from the ER retrieval/retention pathway that is mediated by the interaction of its ER retention motif with Coatomer subunit beta (COPB1).

Participants:
(1) G-protein coupled receptor 15 (GPR15)
(2) 14-3-3 protein beta/alpha (YWHAB)
(3) Coatomer subunit beta (COPB1)

Interactions
Interaction #1 GPR15 - YWHAB

Is mutually exclusive with Interaction #2 GPR15 - COPB1

Interfaces
(1) ELM:LIG_14-3-3_4 motif (356RSVSL360) in G-protein coupled receptor 15 (GPR15)
(2) 14-3-3 protein (5-238) in 14-3-3 protein beta/alpha (YWHAB)

Interaction Regulation
PTM-dependent Induction (Phosphorylation of S359 on G-protein coupled receptor 15 (GPR15)) of the G-protein coupled receptor 15 (GPR15) LIG_14-3-3_4 motif - 14-3-3 protein beta/alpha (YWHAB) 14-3-3 protein interaction

Interaction #2 GPR15 - COPB1

Is mutually exclusive with Interaction #1 GPR15 - YWHAB

Interfaces
(3) TRG_ER_diArg_1 motif (352RRR354) in G-protein coupled receptor 15 (GPR15)
(4) Adaptin N terminal region (18-539) in Coatomer subunit beta (COPB1)

Interaction Regulation
Effector binding Abrogation (14-3-3 protein beta/alpha (YWHAB)) of the G-protein coupled receptor 15 (GPR15) TRG_ER_diArg_1 motif - Coatomer subunit beta (COPB1) Adaptin N terminal region interaction
