Switch #:
SWTI000320
Switch type:
Specificity
Switch subtype:
Motif hiding

Description:
The basic ER retention motif of HLA class II histocompatibility antigen gamma chain (CD74) is blocked from binding to Coatomer subunit beta (COPB1) by phosphorylation-induced binding of 14-3-3 protein beta/alpha (YWHAB), regulating its release from the ER and trafficking to the plasma membrane.

Participants:
(1) HLA class II histocompatibility antigen gamma chain (CD74)
(2) 14-3-3 protein beta/alpha (YWHAB)
(3) Coatomer subunit beta (COPB1)

Interactions
Interaction #1 CD74 - YWHAB

Is mutually exclusive with Interaction #2 CD74 - COPB1

Interfaces
(1) LIG_14-3-3_3 motif (5RSRSCR10) in HLA class II histocompatibility antigen gamma chain (CD74)
(2) 14-3-3 protein (5-238) in 14-3-3 protein beta/alpha (YWHAB)

Interaction Regulation
PTM-dependent Induction (Phosphorylation of S8 on HLA class II histocompatibility antigen gamma chain (CD74)) of the HLA class II histocompatibility antigen gamma chain (CD74) LIG_14-3-3_3 motif - 14-3-3 protein beta/alpha (YWHAB) 14-3-3 protein interaction
