Switch #:
SWTI000337
Switch type:
Specificity
Switch subtype:
Competition

Description:
T-cell surface antigen CD2 (CD2) uses overlapping motifs to bind to CD2 antigen cytoplasmic tail-binding protein 2 (CD2BP2) and Fyn, which makes their interactions mutually exclusive. Since CD2BP2 and Tyrosine-protein kinase Fyn (FYN) reside in different subcellular locations, the specificity of CD2 for the two competitors is switched by changing its cellular localization, from non-raft membranes to lipid raft membranes.

Participants:
(1) T-cell surface antigen CD2 (CD2)
(2) CD2 antigen cytoplasmic tail-binding protein 2 (CD2BP2)
(3) Tyrosine-protein kinase Fyn (FYN)

Interactions
Interaction #1 CD2 - CD2BP2

Is mutually exclusive with Interaction #2 CD2 - FYN

Interfaces
(1) LIG_GYF motif (295HRPPPPGHR303) in T-cell surface antigen CD2 (CD2)
(2) GYF domain (281-338) in CD2 antigen cytoplasmic tail-binding protein 2 (CD2BP2)

Interaction Regulation
Effector binding Inhibition (Tyrosine-protein kinase Fyn (FYN)) of the T-cell surface antigen CD2 (CD2) LIG_GYF motif - CD2 antigen cytoplasmic tail-binding protein 2 (CD2BP2) GYF domain interaction

Additional Information
Structural information: 1L2Z
Interaction #2 CD2 - FYN

Is mutually exclusive with Interaction #1 CD2 - CD2BP2

Interfaces
(3) LIG_SH3_3 motif (294SHRPPPP300) in T-cell surface antigen CD2 (CD2)
(4) SH3 domain (88-135) in Tyrosine-protein kinase Fyn (FYN)

Interaction Regulation
Effector binding Inhibition (CD2 antigen cytoplasmic tail-binding protein 2 (CD2BP2)) of the T-cell surface antigen CD2 (CD2) LIG_SH3_3 motif - Tyrosine-protein kinase Fyn (FYN) SH3 domain interaction
