Switch #:
SWTI000493
Switch type:
Binary
Switch subtype:
Pre-translational

Description:
Alternative splicing inserts exons within the Caspase-3 scission motif of Protein kinase C delta type (PRKCD), abrogating binding to Caspase-3 (CASP3). Cleavage of PKCdeltaI Protein kinase C delta type (PRKCD) by caspase-3 releases a catalytically active C-terminal fragment that is sufficient to induce apoptosis. This inserted exon disrupts scission motifs and therefore the PKCdeltaVIII (GENBANK:DQ516383) splice variant functions as an anti-apoptotic protein in NT2 cells.

Participants:
(1) Protein kinase C delta type (PRKCD)
(2) Caspase-3 (CASP3)

Interactions
Interaction #1 PRKCD - CASP3

Interfaces
(1) CLV_C14_Caspase3-7 motif (326DMQDN330) in Protein kinase C delta type (PRKCD)
(2) Caspase domain (45-275) in Caspase-3 (CASP3)

Interaction Regulation
Alternative splicing Abrogation of the Protein kinase C delta type (PRKCD) CLV_C14_Caspase3-7 motif - Caspase-3 (CASP3) Caspase domain interaction
